Nonspecific T-lymphocyte mitogenesis by pyrogenic exotoxins from group A streptococci and Staphylococcus aureus.

چکیده
Group A streptococcal pyrogenic exotoxins types A, B, and C and staphylococcal pyrogenic exotoxin were shown to be potent nonspecific T-lymphocyte mitogens. Adherent cell populations did not significantly affect the nonspecific mitogenicity.

Ganglioside and monosaccharide inhibition of nonspecific lymphocyte mitogenicity by group A streptococcal pyrogenic exotoxins.
Group A streptococcal pyrogenic exotoxins (SPE) types A, B, and C are potent nonspecific lymphocyte mitogens. The mitogenicity of these exotoxins was inhibited by gangliosides and sialic acid, whereas concanavalin A was unaffected. The capacity of both concanavalin A and SPE-A to stimulate lymphocytes was suppressed by alpha-methyl-D-mannopyranoside. Galactose reduced the activity of SPE-C. The...
متن کاملPurification and properties of an extracellular blastogen produced by group A streptococci
An extracellular product of group A streptococci which induces lymphocyte blastogenesis has been purified to homogeneity by DEAE-cellulose and CM-cellulose chromatography. The protein, termed streptococcal blastogen A, has a mol wt of approximately or equal to 17,500 and is inactivated by protease treatment and by heating at 100 degrees C. The purified blastogen gave rise to multiple protein ba...
متن کاملCytolysins augment superantigen penetration of stratified mucosa.
Staphylococcus aureus and Streptococcus pyogenes colonize mucosal surfaces of the human body to cause disease. A group of virulence factors known as superantigens are produced by both of these organisms that allows them to cause serious diseases from the vaginal (staphylococci) or oral mucosa (streptococci) of the body. Superantigens interact with T cells and APCs to cause massive cytokine rele...
متن کاملA single, engineered protein therapeutic agent neutralizes exotoxins from both Staphylococcus aureus and Streptococcus pyogenes.
Staphylococcus aureus and Streptococcus pyogenes secrete exotoxins that act as superantigens, proteins that cause hyperimmune reactions by binding the variable domain of the T-cell receptor beta chain (Vβ), leading to stimulation of a large fraction of the T-cell repertoire. To develop potential neutralizing agents, we engineered Vβ mutants with high affinity for the superantigens staphylococca...
متن کاملExotoxins of Staphylococcus aureus.
This article reviews the literature regarding the structure and function of two types of exotoxins expressed by Staphylococcus aureus, pyrogenic toxin superantigens (PTSAgs) and hemolysins. The molecular basis of PTSAg toxicity is presented in the context of two diseases known to be caused by these exotoxins: toxic shock syndrome and staphylococcal food poisoning. The family of staphylococcal P...
